Streaking from glycerol stock into LB Agar:
Materials:

Glycerol stock of bacteria
(stored at –80°C)

Sterile LB agar plate
-
Inoculating loop (sterile)
Steps:

Take the glycerol stock out of
the –80°C freezer and keep on ice.

Take a sterile inoculating
loop.

Dip into the glycerol stock (a
tiny scrape is enough).
-
Streak across a quadrant of the
LB agar plate using a zig-zag motion.
-
Take a new sterile loop.
-
Streak from the end of the
first quadrant into a new section (repeat 3-4 times to dilute cells).
-
Invert the plate and incubate
overnight at:
-
37°C (common strains like E.
coli.
-
Check for colonies after 12–18
hours.
